Project 1: Defining the contribution of lipid and glucose metabolism in regenerative angiogenesis

Sirtuin-3 (Sirt3) is a deacetylase that governs fatty acid b-oxidation and mitochondrial energy production. Modulating Sirt3 expression in a mice model of proliferative retinopathy will help us decipher the role of glucose and lipid metabolism as novel determinants of regenerative and pathological angiogenesis. Our exciting pilot results show that Sirt3-deficient mice initiate in vessels a regenerative program that is dependent on aerobic glycolysis. We hypothesize that a shift in energy metabolism towards aerobic glycolysis improves retinal vascular regeneration and neuronal function.
